Joe, The curing is a chemical reaction that takes a while to complete. 
24 to 48 hours should be enough before sealing.  If you want, place it
in an oven at a low temperature (160 degrees) after 48 hours and bake it
for an hour or two.  You don't want a high temperature (over 212
degrees) because any residual water will flash to steam and explode.

Mutalib,  I have sealed my plaster tools with epoxy, mainly because I
had several gallons given to me years ago.  Any good paint will do as
long as the tool doesn't sit in a pool of water for a long time. 
Painting makes it easy to seal after every grit.  I thing Mel suggests
using a thinned pitch for sealing.  

Wax around the edges and on the bottom will work too.  Tom Waineo used
parafin wax to set his tiles on the tool and as a sealant.  Can't get
much metter waterproofing than that, and it makes it easy to change
tiles if necessary.
