Like many others here I use Vuescan to capture 48bit tiff files, with no
color correction, and then adjust curves and levels with Photoshop. One
feature that's handy for astroslides with Nikon scanners is that you can
adjust the analog gain for very dark slides.. results will be noisy but can
be probably compensated with multi-scanning (another Nikon feature)..
Vuescan will allow manual focus but your problem is a narrow focus field in
the Nikon.. perhaps you can solve this by mounting the slide in a glass
mount.. that should keep the slide flat and focused.
Vuescan does support dust/scratch removal using the infrared channel, also
single pass multiscanning and manual focus. I use it in the auto mode to
scan family pictures, very good and very fast...
