AIP4WIN is all you need: that and two flats, one bias and one dark of the 
same exposure time as the flats. Make sure they are all at the same 
temperature.

In the AIP4WIN version 1.4.25, under the Measure Menu item, is a 
"CHARACTERIZE CCD CAMERA" command. Just select that and follow the prompts

Run the camera cold because the read noise is a function of temperature.

Target the flats to be about 50% of the ADU range. MY IMG6303 has a bias of 
about 3000 counts so I try to set my flats at about 35K roughly. It isn't 
very critical. But try to have nearly identical flats.
